DNA test for the MFSD8 c.843delT variant that causes NCL7 in Chihuahua and Chinese Crested dogs.
Overview
This genetic test analyses the MFSD8 c.843delT variant in Chihuahua and Chinese Crested dogs. The condition is known as neuronal ceroid lipofuscinosis 7, NCL7, CLN7, a Batten-like neurodegenerative disorder and MFSD8-related lysosomal storage disease.
MFSD8 encodes a lysosomal membrane/transport protein. The deletion causes a frameshift with an early stop codon, strongly truncating the protein and allowing storage material to accumulate in nervous tissue.
NCL7 is a severe progressive neurodegenerative disorder. Affected dogs can develop blindness, anxiety, disorientation, compulsive behaviour, increased sleeping, sensitivity to stimuli, tremors, seizures and loss of coordination at a young age.
NCL disorders are lysosomal storage diseases in which nerve cells are gradually damaged. As a result, problems with vision, behaviour, coordination, movement and neurological function can increase over time.
The trait is inherited as autosomal recessive. Two copies of the variant cause the condition. Carriers have one copy, usually appear healthy, and can pass the variant on.
For breeders, this is especially useful because carriers can look healthy while two carriers together can produce affected puppies.
Included subanalyses
This analysis includes the following subanalysis:
Allele combinations & result interpretations
Below, for each tested locus, you will find the possible allele combinations that may be reported within this analysis, together with a brief explanation of their genetic meaning. The interpretation of possible interactions between different loci is included in the report, but is not shown here in full because that would lead to too many combinations on this page. The final expression may also depend on other genes and their interaction.
Genotype / allele combination: Clear (N/N)
The dog does not carry the tested MFSD8 c.843delT variant. This animal will not develop MFSD8-related NCL7 due to this specific variant and will not pass it on.
Genotype / allele combination: Carrier (N/delT)
The dog carries one copy of the MFSD8 c.843delT variant. This animal is a carrier, is generally not affected by one copy, and can pass the variant on.
Genotype / allele combination: Affected genotype (delT/delT)
The dog carries two copies of the MFSD8 c.843delT variant. This genotype causes MFSD8-related NCL7.
