DNA test for the ADAMTSL2 c.661C>T variant that causes Musladin-Lueke syndrome (MLS) in Beagles.
Overview
This genetic test analyses the ADAMTSL2 c.661C>T variant in the Beagle. The condition is known as Musladin-Lueke syndrome, MLS, Chinese Beagle syndrome, Ballerina Beagle syndrome and ADAMTSL2-related connective tissue disorder.
ADAMTSL2 is involved in extracellular matrix and connective tissue development. The c.661C>T variant alters the protein and causes an inherited disorder in Beagles that can clearly affect skin, joints, growth and posture.
Affected Beagles can have tight or stiff skin, a compact body, restricted joint movement, abnormal gait and a characteristic “ballerina” stance with walking high on the toes. A broad skull, slanted eyes, folded ears, shortened outer toes and a tense facial expression are also described. Severity varies, but the genotype is important because carriers can look normal.
MLS is inherited as autosomal recessive. Two copies of the variant cause the condition. Carriers have one copy, usually appear healthy, and can pass the variant on.
Because the full appearance of MLS may become clearer over time and carriers can be clinically normal, DNA testing gives a direct and practical basis for responsible Beagle breeding decisions.

Genotype / allele combination: Carrier (CT)
The dog carries one copy of the ADAMTSL2 c.661C>T variant. This animal is a carrier, is generally not affected by this single copy, and can pass the variant on.
Genotype / allele combination: Affected genotype (TT)
The dog carries two copies of the ADAMTSL2 c.661C>T variant. This genotype causes Musladin-Lueke syndrome in the Beagle.
