Overview

What does this test examine?

This genetic test analyses the ADAMTSL2 c.661C>T variant in the Beagle. The condition is known as Musladin-Lueke syndrome, MLS, Chinese Beagle syndrome, Ballerina Beagle syndrome and ADAMTSL2-related connective tissue disorder.

ADAMTSL2 is involved in extracellular matrix and connective tissue development. The c.661C>T variant alters the protein and causes an inherited disorder in Beagles that can clearly affect skin, joints, growth and posture.

What does this condition mean?

Affected Beagles can have tight or stiff skin, a compact body, restricted joint movement, abnormal gait and a characteristic “ballerina” stance with walking high on the toes. A broad skull, slanted eyes, folded ears, shortened outer toes and a tense facial expression are also described. Severity varies, but the genotype is important because carriers can look normal.

Inheritance and result

MLS is inherited as autosomal recessive. Two copies of the variant cause the condition. Carriers have one copy, usually appear healthy, and can pass the variant on.

What does the result mean?

  • Clear (C/C): the tested variant was not detected.
  • Carrier (C/T): the dog carries one copy and can pass the variant on.
  • Affected (T/T): two copies cause the MLS condition.

Practical value of this test

  • The test identifies hidden carriers before Beagles are used in breeding combinations.
  • Breeders can avoid carrier-to-carrier matings and prevent puppies with MLS.
  • The result helps manage valuable carriers responsibly without unnecessarily excluding lines.
  • In Beagles with unusual build, tight skin or typical gait, the result supports a clear hereditary explanation.
  • The test gives customers concrete information for mating plans, puppy selection and transparent communication with buyers.

Because the full appearance of MLS may become clearer over time and carriers can be clinically normal, DNA testing gives a direct and practical basis for responsible Beagle breeding decisions.
