DNA test for the SLC27A4 c.1250G>A variant causing autosomal recessive congenital ichthyosis in Great Danes.
Overview
This genetic test analyses the SLC27A4 c.1250G>A variant in Great Dane. The disorder is known as Ichthyosis Great Dane type / congenital ichthyosis, ichthyosis, congenital ichthyosis and an inherited cornification disorder. The name ichthyosis refers to dry, scaly skin that can resemble fish scales.
SLC27A4 encodes a fatty-acid transport protein important for skin-barrier lipids. The c.1250G>A variant alters splicing and reduces normal protein function in the skin. A disrupted skin barrier can lead to scaling, dry or thickened skin, a dull coat, redness, itching, fissures or secondary skin infections. Severity differs between types and breeds: some forms are mainly chronic and inconvenient, while others cause serious skin disease in young puppies. Affected Great Dane puppies can show severe generalized hyperkeratosis with strongly wrinkled, thickened and scaling skin, especially around the eyes and nose.
The condition is inherited as an autosomal recessive trait. One copy makes a dog a carrier; two copies cause the severe Great Dane form of ichthyosis.
Included subanalyses
This analysis includes the following subanalysis:
Allele combinations & result interpretations
Below, for each tested locus, you will find the possible allele combinations that may be reported within this analysis, together with a brief explanation of their genetic meaning. The interpretation of possible interactions between different loci is included in the report, but is not shown here in full because that would lead to too many combinations on this page. The final expression may also depend on other genes and their interaction.
Genotype / allele combination: Clear / normal genotype (G/G)
The tested SLC27A4 variant was not detected. This dog will not develop SLC27A4-gerelateerde ichthyosis from this variant and will not pass it on.
Genotype / allele combination: Carrier (G/A)
The dog carries one copy of the tested SLC27A4 variant. Carriers are usually not affected themselves but can pass the variant on; use this result actively in breeding plans.
Genotype / allele combination: Affected genotype (A/A)
The dog has two copies of the tested SLC27A4 variant. This genotype causes SLC27A4-gerelateerde ichthyosis in the relevant breeds and explains the genetic basis for the scaling skin disorder.
